It was City's second defeat in three games and saw them surrender top spot in Division One South and West of the Southern League.
And McCarthy is determined there will be no repeat of City's substandard display at Uxbridge.
"There will be a lot of changes for Saturday," he said.
"I think only keeper Martin Rice, who had a lot to do, came out of the game with any credit.
"Too many players went missing. The performance was unacceptable."
And McCarthy also revealed he is actively looking to strengthen the squad.
"I am looking at two or three players though it might be too soon to have them here by Saturday."
What angered McCarthy most was his side's lack of discipline.
"We fell into the oldest trick in the book. They were in our face and wound us up and we ended up arguing amongst ourselves and the other team were laughing at us.
"They wanted it more than us which was one of the most disappointing things.
"I am honest and I ask the players to be honest but I did not get it on Saturday."
McCarthy and coach Dave Newton called a meeting with the players ahead of Tuesday evening's training session.
"I can't repeat what I am going to tell them but they will be getting a few home truths.
"I just did not see that performance coming. There are no excuses. We have got to stop these Jekyll and Hyde displays."
Saturday's game is one City will be expected to win comfortably against a Winchester side who have taken just seven points out of a possible 45 this season and have only one win to their name.
On Tuesday evening they drew 1-1 at home to Thatcham Town.
And anything less than a convincing victory will set the alarm bells ringing at Treyew Road.
It is the start of a run of four home league games in their next five matches culminating in an FA Vase rematch against AFC Totton December 9.
It is essential City put together a good run of results to stay on track for a fourth successive promotion.
